Since his PhD in the lab of Piet Borst (1980), Johannes (Hans) Bos has studied the molecular cause of cancer. Initially in the lab of Alex van der Eb in Leiden, using adenovirus 12, a virus that can form tumors in rodents. In 1984 he started to work on Ras mutations in human tumors. He could demonstrate that such mutations occur frequently in many tumor types. After a sabbatical year in San Francisco with Frank McCormick he moved to Utrecht in 1991 to become professor in Molecular Cancer Research and to work on the function of the Ras protein. In 1994 he started to work on the small GTPase Rap, leading to the discovery of the cAMP target Epac. More recently, he joined forces with Hans Clevers, to study Ras in patient-derived tumor organoids. From 2007-2015 he was chair of the Divison Biomedical Genetics and head of the Department Molecular Cancer Research. Currently he chairs of the UMCU priority program Cancer. In 2014 he received the prestigious Josephine Nefkens prize for Cancer Research.

Current research focuses on three topics:
1. The function of the small GTPase Rap1 and the cAMP regulated guanine nucleotide exchange factor Epac in the control of cell adhesion.
2. The mechanism of brush border formation and apical membrane formation in intestinal epithelial cells
3. Targeted therapies for mutant Ras containing colorectal cancer
